This series of works is focused on investigation of molecular components of cell associated with neurodegenerative diseases such as Alzheimer’s disease, Huntington’s disease, as well as Down syndrome and Saethre-Chotzensyndrome.

A few fundamental results related to structural and functional diversity of important regulator of endocytosis and signaling intersectin 1 were obtained. Intersectin 1 is implicated in neurodegenerative processes during Alzheimer’s and Huntington’s diseases, as well as Down syndrome. Precise mode of posttranscriptional regulation of Twist gene linked to Saethre-Chotzensyndrome was determined.

This work resulted into priority data acquisition that are important for our understanding of molecular interactions and networks of gene regulation of cellular components involved into development and progression of neurodegenerative diseases.

Two novel classes of inhibitors of protein kinase ASK1, which regulates neuron death during several neurodegenerative pathologies, were identified. These compounds increased survival rate of cells during apoptosis. Discovered inhibitors could be used as a tool for investigation of ASK1 role in mentioned diseases as well as in long-term outlook for combined therapy of neurodegenerative pathologies. One of the inhibitors of   ASK1 – NQDI-1 is already commercially available at the catalog of Sigma Aldrich corporation.

Obtained data are important for medical diagnostics and therapy of neurodegenerative diseases. Moreover these results could be used during investigation of cancer types related to aberrant intracellular signaling.
