Aim of the study: to identify the molecular and genetic changes in radiation-induced (post-Chornobyl) and sporadic papillary thyroid carcinoma and to determine the diagnostic and prognostic role of these changes; to identify and verify proteins as persepctive for preoperative diagnosis markers of papillary thyroid carcinoma.
In the research work the data about specific molecular changes were obtained (rearrangements RET/PTC1, RET/PTC3, BRAF 1799T>A mutation; expression of Ki-67 (MIB-1 index), BCL-2, cyclin A, cyclin D1, status amplification of cyclin D1) and clinical features of post-Chornobyl papillary cancer in adults were investiagted in relation to other cohorts. The role of the gene encoding telomerase reverse transcriptase(TERT) for malignant transformation of thyroid cancer cells was investigated. The TERT promoter mutation 228C> T and 250C> T were investigated in post-Chornobyl papillary thyroid carcinoma.
By a proteomic approach a protein S100A6 protein was identified and verified as a diagnostic marker of papillary thyroid cancer. Also using a proteomics a proteins 14-3-3, PRX6, ANXA5, SELEBP1, PDIp were identified with a different expression levels in follicular thyroid tumors and papillary thyroid carcinoma and the role of these protein was tested for differential diagnosis of bening and malignant thyroid tumors.
A cystic fluid to be accumulated in benign thyroid cysts and cystic papillary thyroid carcinoma was evaluated by using a proteomic approach. A role of protein S100A13 and cytokeratin 19 (CK-19) was evaluated and verified as candidate protein markers for cystic papillary thyroid carcinoma. The first extensive catalogue of the protein content in fluid from the thyroid cystic neoplasms was created, which has a clinical significance and expands the understanding of the carcinogenesis of these tumors.
